Recombinant monoclonal antibody to Human IL1 beta. Canakinumab is a human monoclonal antibody targeted at interleukin-1 beta. It has no cross-reactivity with other members of the interleukin-1 family, including interleukin-1 alpha.
Canakinumab was for the treatment of rheumatoid arthritis. Canakinumab is as a possible treatment for chronic obstructive pulmonary disease, gout and coronary artery disease.

    The article investigates the mechanism by which the SARS-CoV open reading frame 3a (ORF3a) protein activates the NLRP3 inflammasome, contributing to the cytokine storm characteristic of severe coronavirus infections. The researchers demonstrate that ORF3a uniquely activates both signals required for inflammasome activation: pro-IL-1β gene transcription through NF-κB activation, and IL-1β protein maturation through ASC-dependent caspase-1 activation. They identify a TRAF binding domain in ORF3a that enables interaction with TRAF3, which mediates K63-linked ubiquitination of ASC and promotes ASC speck formation. Unlike other viral inflammasome activators, ORF3a's ability to stimulate inflammasome activation is independent of its ion channel activity but requires TRAF3, NLRP3, and ASC.
    Creative Biolabs provided Canakinumab, a therapeutic antibody that blocks IL-1β, which was used to assess the impact of IL-1β inhibition on SARS-CoV replication. The researchers demonstrated that treatment with this antibody mildly increased viral replication, suggesting that inflammasome activation has a weak suppressive effect on SARS-CoV. This finding helps clarify the dual role of inflammasome activation in coronavirus infections - serving both antiviral functions and potentially contributing to pathological inflammation - providing valuable insights for therapeutic strategies targeting coronavirus-induced inflammatory responses.
